No, you do not need to uninstall the previous version to install the new one. 

 

The Introductory Tutorial located here <http://doe2.com/download/equest/eQ-v3-64_Introductory-Tutorial.pdf>  addresses this, please see section: Installing eQUEST on pp 4 of 145 (pp 10 of the pdf). It also provides guidance regarding the installation in different Windows operating systems (Vista and XP).
