Yes.  

If you want to simulate just one day, you can use the DesignDay object and 
specify the hourly temperature profile using the fields for "Dry-Bulb 
Temperature Range Modifier Type" and "Dry-Bulb Temperature Range Modifier 
Schedule".

If you want to simulate more than one day using a weather file, you can take a 
weather file for the location of interest and edit it.  Using the EnergyPlus 
Weather Converter, you can convert an epw file to a special csv format which 
contains column headings to identify the data and is easily editable in a 
spreadsheet program such as Excel.  After editing, save (still in csv format) 
and then use the weathter converter to convert from csv to epw.  When using 
this approach, you will also need to make sure that the dew point and relative 
humidity data are compatible with the dry bulb temperatures, and make changes 
to any other weather data columns as appropriate.  The weather converter can be 
accessed from the Start Menu (on windows systems):

Start Menu\Programs\EnergyPlus V2-1 Programs\Weather Statistics and Conversions

or directly from the file explorer at

C:\EnergyPlusV2-1-0\PreProcess\WeatherConverter\Weather.exe
