Before tackling specific questions such as this, I advise spending time to first familiarize yourself with the software by making a simple model.  Start up the program, choose to  create a new project, select "design development wizard," then invest your time exploring all the input screens and windows available for the wizard mode.  As an exercise, build a simple model of the building you work in or any simple building you are familiar with, and spend a moment to consider how each input applies to creating that model before moving on to the next screen.  Notice some screens are unavailable until you choose to use a certain system or choose to use optional inputs for a shell.

On finishing the above, you will be prepared to leverage the wizards for simple models and studies, and ready to approach learning detailed mode edits - which make some wizard-level things easier and are necessary to leverage the larger portion of what this software can accomplish.

To answer your follow up directly, you can define U-values for constructions both in wizard mode and in detailed mode.  Cheng shared a screenshot of one means of doing this in the detailed mode (another is in the spreadsheet view).  You get to either by navigating to Building Shell tab, selecting the construction in the component tree to the left (you'll probably need to scroll down a bit), then either make the edit in spreadsheet view - easier when you need to make the same change to many components, or doubleclick the component to make the change in the input window Cheng showed.

How you define the construction can be important - search the archives or read the associated help entries to understand the differences between constructions defined only by U-value and those defined with layers.  I generally use layers and edit one or more layers to achieve a target U-value.

The alternative is to make the construction edits in the wizards, within the shell input screens.  You can choose to either generally specify the layers of the construction, or to define a custom construction, again either in layers and/or with an specified overall R-value.  Same cautions about non-layered constructions apply from above.  Following show the DD wizard input screens I'm discussing:
