No -- the sizing calculations are done over a 24 hour period (for the Design Day) or whatever period (Weather file sizing period input) and use the critical/peak conditions for your building to calculate the appropriate sizes of supply air or components.

No -- Don't run the simuation for Sizing Periods means you will not have any results shown for the Output:Variable or Output:Meter objects you might have put in.  The sizing will still be done using the 24 hour period "described" by or selected by the objects.
