but  for example if i set the PlantLoop Supply inlet Node to maintain a certain temperature,  then what happens ?

A SetpointManager is basically a control point - it indicates the status to be maintained at that point in the loop.  For example, if you set the PlantLoop Supply Outlet Node to maintain a certain temperature, the equipment on the supply side of the plant loop will need to operate to maintain that temperature.  If the PlantLoop is a hot water loop and there is a boiler on the supply side, then the boiler might modulate to maintain the setpoint temperature.  Not all equipment is capable of maintaining all setpoints, so sometimes the setpoint is not met and other problems can result, but think about it as a desired state within the plantloop controls.
