No, it only matters that placing a set point here will not conflict with another set point "at this same node". Since you don't need a set point here to control the fan, using this node as a place holder for a set point "that will be used by a mixed air set point manager" is fine.
2) Should I insert a Duct object as the last componant on the supply side, before opting for the exhaust node (this may be important for DOAS).
You don't need that, there are plenty of uncontrolled air nodes in a simulation.
